Intended Uses: This immunoassay kit allows for the quantitative in vitro determination of general concentrations of CdiGMP in serum, plasma, tissue homogenates, cell culture supernatants, and other biological fluids.
Principle of the assay: The ELISA is based on the competitive binding enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with an antibody specific for CdiGMP; During the reaction, CdiGMP in the sample or standard competes with a fixed amount of biotin-labeled CdiGMP for sites on a pre-coated antibody specific for CdiGMP. Excess conjugate and unbound samples or standards are washed from the plate.
Avidin conjugated to horseradish peroxidase (HRP) is then added to each well of the microplate and incubated. A TMB Substrate Solution is then added to each well. The enzyme-substrate reaction is terminated by adding a sulfuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of cGMP in the samples is determined by comparing the O.D. of the samples to the standard curve.
